ABSTRACT

OBJECTIVE:To investigate the correlation between the morphological characteristics and chemical constituents of garden ginseng (GG),mountain transplanted ginseng (MTG)and mountain-grown ginseng (MGG),so as to provide evidence for elucidating the scientific connotation of “evaluating quality from morphological characteristics ”. METHODS :A total of 30 batches of GG ,MTG and MGG were collected. The contents of 11 saponins(ginsenoside Rg 1,Re,Rg2,F1,Rb1,Rb2,Rb3,Rc,Rd,Ro, Rf)were determined by UPLC. The determination was performed on Acquity UPLC ® BEH C 18 column with mobile phase consisted of 0.1% Formic acid solution- 0.1% Acetonitrile formate solution (gradient elution )at the flow rate of 0.5 mL/min. The column temperature was set at 35 ℃. The detection wavelength was set at 203 nm,and sample size was 5 μL. The content of starch was determined by colorimetry ,and the absorbance value was measured at 559 nm with 0.5% iodine-potassium iodide as the color reagent. SPSS 23.0 statistical software was used to perform Pearson relationship analysis for appearance characteristic indexes (number of stem scars ,rhizome length ,texture)with the contents of 11 kinds of saponins ,the content of total saponins ,the content ratio of panaxatriol saponin (PPT)/panaxadiol saponin (PPD),the content of starch. RESULTS :The linear relationship between UPLC and colorimetry was good (r>0.999). RSDs of precision ,reproducibility and stability tests were all lower than 5% . Average recoveries were 95.17% -105.20%(RSD<5% ,n=6). Except for ginsenoside Rf ,the contents of other 10 ginsenosides were positively correlated with the number of stem scars ,so were the contents of 11 gensenosides with the rhizome length(r>0,P<0.05),and they were negatively correlated with the firmness of the texture (r<0,P<0.05). The content of starch in P. ginseng was positively correlated with texture firmness (r=0.95,P<0.01). The ratio of PPT/PPD in P. ginseng was negatively associated with the number of stem scars and the rhizome length (r<0,P<0.05). CONCLUSIONS :The number of stem scars ,the rhizome length and texture porosity of P. ginseng are positively associated with the contents of medicinal material ginsenoside;the increase of PPD content in P. ginseng was higher than that of PPT with the increase of growth years. The traditional“evaluating quality from morphological characteristics ”for P. ginseng has a certain scientific connotation ,which is more stem scars ,longer rhizome and loose texture.

ABSTRACT

Objective The incidence of complications after esophageal cancer surgery is relatively high.The purpose of this paper was to explore the feasibility of combined thoraco-laparoscopy surgical treatment for senile esophageal cancer(over 70 years old).Methods A total of 526 esophageal cancer cases were retrospectively analyzed in this study.The operative procedures was esophageal carcinoma resection surgery and thoracic and abdominal lymphadenectomy which were operated through video-assisted by thoracoscope and laparoscopic From January 2010 to June 2014 in the Union Hospital of Fujian Medical University,divided into aged groups(≥70 years old,n =132) and non-aged group(< 70 years old,n =394).Statistical analysis was carried on the rate of two groups of preoperative risk factors(hypertension,diabetes,cardiac insufficiency,pulmonary insufficiency,cerebral infarction),surgical trauma and the number of lymph node dissection (laryngeal recurrent nerve injury,operation time,intraoperative blood loss),postoperative complications(cardiac complications,chylothorax,pulmonary complications,anastomotic fistula,poor healing of the incision,intrathoracic stomach atony),postoperative recovery and early postoperative mortality.Results Preoperative risk factors:the prevalence of the aged group with hypertension,cerebral infarction,cardiac insufficiency,and pulmonary insufficiency was significantly higher than the non-aged group (P < 0.05) but diabetes (P > 0.05).Surgical trauma and the number of lymph node dissection:no significant difference (P > 0.05).Postoperative complications:the aged group was higher in the incidence of pulmonary complications and cardiac complications than the other (P < 0.05) ; but the chylothorax,anastomotic fistula and poor healing of incision,intrathoracic stomach atony in the two groups seemed no significant difference(P > 0.05).Furthermore,pulmonary complications were highest in the all complications,significantly higher than the others in both groups.Postoperative recovery:significant difference existed in postoperative hospitalization days,the aged group was obviously longer than the non-age groups.Early postoperative mortality rates in the two groups had no significant difference.Conclusion The aged patiences had a higher prevalence in the preoperative risk factors and so was the cardiac complications as well as pulmonary complications among the postoperative complications.However the combined thoraco-laparoscopy in the surgical treatment of elderly patients with esophageal cancer is safe and feasible.

ABSTRACT

BACKGROUND: Reference procedures are required for evaluating the accuracy of routine analytical systems for uric acid (UA). External quality assessment (EQA) for UA has only been conducted with quality controls in China, and the results have not been published. This study was designed to investigate both the trueness and inter-laboratory precision of UA measurements among routine analytical systems using a candidate reference method. METHODS: We performed the HPLC method recommended by the Japan Society of Clinical Chemistry (JSCC). Next, we evaluated its analytical performance and validated its trueness. The performance of 4 routine analytical systems (5 instruments per system, n=20) for UA was assessed by using 4 frozen pooled serum samples measured by the HPLC method according to biologically relevant quality goals. RESULTS: Within-run, between-run, inter-day, and total CV of the method were less than 0.3%, 0.4%, 1.8%, and 2.6%, respectively. The UA measurements were consistent with the target values of standard reference material (SRM) 909b, the sixth ring trial for Reference Laboratories (RELA-2008) specimen, and national primary reference materials. The 4 frozen pooled serum samples were homogeneous, stable, and commutable. All routine systems achieved the desirable performance goal (total error <11.9%). CONCLUSIONS: We successfully reproduced the JSCC's HPLC method, which was simple, specific, precise, and accurate. We recommend this method as a reference method for UA measurement in human serum. Four routine analytical systems for UA measurement had acceptable traceability, and their UA results showed good concordance.

ABSTRACT

Objective To compared two classical rat models of nephrotic syndrome and to provide some reference data to researchers.Methods Thirty male SD rats were randomly divided into control group,puromycin aminonucleoside-induced nephrotic syndrome (PAN) group and adriamycininduced nephrotic syndrome (ADR) group.The body weight,twenty four hour proteinuria level,serum albumin concentration,cholesterol concentration,creatinine and urea concentration were measured.The renal pathology change was evaluated.The drug toxic effects,administration methods and the costs were also compared.Results There was no significant difference in body weight and hair color between control group and PAN group.Compared to control group,the body weight of the rats significantly decreased at day 15 and day 21 in ADR group (P < 0.01),accompanied by epilation and diarrhea.Compared to control group,the 24-hour urinary protein levels increased significantly at day 10 (P < 0.01),day 15 (P < 0.01),and reached the peak level at day 15 (P < 0.01),day 21 (P < 0.01) in PAN group and ADR group respectively.Compared to control group,the serum albumin concentration decreased significantly at day 10 (P<0.01),and return to normal level at day 15.The serum cholesterol concentration was increased significantly at day 10 (P < 0.01) and return to normal at day 15 in PAN group.Compared to control group,the serum albumin concentration was decreased significantly at day 15 (P<0.05) and return to normal at day 21 in ADR group.No significant difference of serum creatinine and serum urea nitrogen levels were found among three groups.Compared to control group,the width of foot process increased significantly at day10 (P < 0.01) and day 15 (P < 0.05) in PAN group and ADR group respectively.To successfully induce a nephrotic rat model (per 100 g),the cost of PAN group was 3.1 times of ADR group (578.10 yuan vs 186.94 yuan).Conclusions Nephrotic syndrome can be induced by both PAN and ADR.The administration of PAN via intraperitoneal injection is more convenient as compared to ADR via tail intravenous injection.Compared to ADR,PAN can induce nephrotic syndrome model more rapidly,with more consistent detection index,and less toxic effects,but its cost is more expensive.

BACKGROUND: Commutable reference materials (RMs) are suitable for end-users for evaluating the metrological traceability of values obtained using routine measurement systems. We assessed the performance of 6 routine measurement systems with validated secondary RMs. METHODS: We tested the homogeneity, stability, and commutability of 5 minimally processed human serum pools according to the standard guidelines. The serum pools were assigned values as per the reference procedure of the United States Centers for Disease Control and were used to evaluate the trueness of results from 6 commercial measurement systems based on enzymatic methods: 3 glucose oxidase (GOD) and 3 hexokinase (HK) methods. RESULTS: The prepared RMs were validated to be sufficiently homogenous, stable, and commutable with the patient samples. Method bias varied for different systems: GOD01, -0.17 to 2.88%; GOD02, 1.66 to 4.58%; GOD03, -0.17 to 3.14%; HK01, -3.48 to -0.85%; HK02, -3.83 to -0.11%, and HK03, -1.82 to -0.27%. CONCLUSIONS: We observed that the prepared serum glucose RMs were qualified for trueness assessment. Most of the measurement systems met the minimal quality specifications.

ABSTRACT

Objective To develop an improved enzymatic method for assaying creatinine in serum and urine. Methods Cratinine iminhydrolase catalyzes the conversion of creatinine to N-methylhydantoin and ammonia,the later combines with 2-oxoglutarate and NADPH in the presence of glutamate dehydrogenase.Before beginning the reaction sequence,endogenous ammonia is removed by an “ammonia scavenger system”that involves the same auxiliary reaction.The NADPH and 2-Oxoglutarate consumed are restored through a reaction catalyzed by isocitrate dehydrogenase.The activity of this strictly magnesium-dependent enzyme is completely blocked by magnesium-complexing reagent that acts promptly whenever the reaction has started with creatinine iminohydrolase as startor.The reaction is monitored kinetically by measuring the NADPH decrease via its absorbance at 340nm.Results The method affords a simple,rapid,and sensitive assay with good precision and extended linearity.Test results compare closely with the HPLC precedure.The proposed method is not subject to interference from several metablites or from the drugs used commonly in clinics.Conclusion The method is easily automated and is suitable for routine work in clinical laboratories.

ABSTRACT

Objective To compare the effect of Rupixiao Tablets prepared by or not by optimized process on hyperplasia of mammary glands in rats.Methods Rat models with hyperplasia of mammary glands were established by injection of estradiol benzoate and progesterone.After treatment with Rupixiao Tablets for four weeks,the mammary gland tissues were sliced for pathological examination,uterine index and ovarian index were measured,the serum contents of estradiol, progesterone,testosterone,prolactin,luteining hormone and follicle stimulating hormone were determined by radioim- munoassay method.Results Compared with the model group,the symptom of red and swollen nipples was relieved,hy- perplasia of lobule of mammary gland and gland vacuole and duct were reduced,the levels of estradiol,progesterone, testosterone,luteining hormone were reduced and prolactin was increased in all treatment groups.Rupixiao Tablets pre- pared by two kinds of process both had evidently therapeutic effect,and the differences of the compared indexes were in- significant.Conclusion Rupixiao Tablets prepared by optimized process has therapeutic effect on hyperplasia of mamma- ry glands in rats and has no significant differences in the main pharmacodynamics indexes when compared to those of the original preparation.

ABSTRACT

Objective To establish the determination of borneol and menthol in C ompound Shexiang Injection by HS-GC.Methods A Rtx-Wax column (30 m?0.25 mm? 0.25 ?m) and FID were used.The injector and the detector temperature was 260 ℃ .Column temperature was controlled by programming:initial temperature at 140 ℃and holding 2 min,and temperature increase at 10 ℃?min-1,final temperatur e at 220 ℃for 4 min. Results The linear range of borneol and menthol was 13.025 ~416.8 ?g?mL-1 (r=0.999 5,n=6) and 13.125~420 ?g?mL-1 (r=0.999 7,n=6) ,and the average recovery was 98.97 %(RSD=3.0 %,n=6) and 97.65 %(RSD=2.1 % ,n=6),respectively.Conclusion The method is sensitive,accurate and reproduci ble for simultaneous determination of borneol and menthol in Compound Shexiang I njection.

ABSTRACT

AIM:To establish the simultaneous determination of menthol, borneol, patchoulic alcohol and muscone in Compound Shexiang Injection (Moschus, Herba Pogostemonis, Borneolum Syntheticum, Mentholum, etc) by GC. METHODS: The GC system consisted of capillary column, Rtx-Wax-30 M; FID as the detector. The programmed temperature-GC and internal standard method were employed to determine four kinds of components in Compound Shexiang Injection. RESULTS: Menthol, borneol, patchoulic alcohol and muscone in Compound Shexiang Injection and n-octadecane have been separated well under the same chromatographic condition. The average recoveries of menthol, borneol, patchoulic alcohol and muscone were 99.76% (RSD=1.48%), 99.36%(RSD=1.99%), 99.15%(RSD=1.67%), 98.87%(RSD=1.57%), respectively. CONCLUSION: The method is sensitive, accurate, separable and reproducible, and can be used to control the quality of Compound Shexiang Injection.
